Some help with spraying

Im in the process of spraying my car and im doing the back bumper.. Filled and sanded and all that but i noticed i missed some filling.. Can i put some isopon on it and give it another coat of primer or do i need to sand the primer off the area that needs filled then prime again? Thanks
